Antigenotoxicity and antimutagenicity of ethanolic extracts of Brazilian green propolis and its main botanical source determined by the Allium cepa test system Genet. Mol. Biol.
Roberto,Matheus Mantuanelli; Jamal,Cláudia Masrouah; Malaspina,Osmar; Marin-Morales,Maria Aparecida.
Abstract Brazilian green propolis is a resinous substance prepared by bees from parts of the plant Baccharis dracunculifolia. As it possess several biological properties, this work assessed the cytotoxic/anticytotoxic, genotoxic/antigenotoxic and mutagenic/antimutagenic potential of ethanolic extracts of Brazilian green propolis (EEGP) and of B. dracunculifolia (EEBD), by means of the Allium cepa test system. The effects were evaluated by assessing the chromosomal aberrations (CA) and micronuclei (MN) frequencies on meristematic and F1 generation cells from onion roots. Chemical analyses performed with the extracts showed differences in flavonoid quality and quantity. No genotoxic or mutagenic potential was detected, and both extracts were capable of...

Antiproliferative and genotoxic effects of Mikania glomerata (Asteraceae) Biocell
Dalla Nora,Gracieli; Pastori,Tamara; Dail Laughinghouse IV,Haywood; Do Canto-Dorow,Thais Scotti; Bosio Tedesco,Solange.
Mikania glomerata is a plant used in Brazilian traditional medicine, known as ‘guaco'. It possesses anti-inflammatory properties and the aqueous extracts of its leaves are indicated for the treatment of diseases of the respiratory tract. This study aimed at evaluating the antiproliferative and genotoxic effect of Mikania glomerata leaf infusions on the cell cycle of onion. The material used was collected in the native environment from Rio Grande do Sul State, Brazil. Aqueous extracts through infusions were prepared in two concentrations: 4g/L (usual concentration) and 16g/L (4x more concentrated) of each of the populations. Two groups of four onion bulbs for each plant population were used plus a control group. The rootlets were fixed in...

Effects of Jatropha curcas oil inLactuca sativa root tip bioassays Anais da ABC (AABC)
ANDRADE-VIEIRA,LARISSA F.; BOTELHO,CAROLINA M.; LAVIOLA,BRUNO G.; PALMIERI,MARCEL J.; PRAÇA-FONTES,MILENE M..
Jatropha curcas L. (Euphorbiaceae) is important for biofuel production and as a feed ingredient for animal. However, the presence of phorbol esters in the oil and cake renders the seeds toxic. The toxicity ofJ. curcas oil is currently assessed by testing in animals, leading to their death. The identification of toxic and nontoxic improved varieties is important for the safe use of J. curcas seeds and byproducts to avoid their environmental toxicity. Hence, the aim of this study was to propose a short-term bioassay using a plant as a model to screen the toxicity of J. curcas oil without the need to sacrifice any animals. The toxicity of J. curcas oil was evident in germination, root elongation and chromosomal aberration tests in Lactuca sativa. It was...

Effects of melatonin on DNA damage induced by cyclophosphamide in rats BJMBR
Ferreira,S.G.; Peliciari-Garcia,R.A.; Takahashi-Hyodo,S.A.; Rodrigues,A.C.; Amaral,F.G.; Berra,C.M.; Bordin,S.; Curi,R.; Cipolla-Neto,J..
The antioxidant and free radical scavenger properties of melatonin have been well described in the literature. In this study, our objective was to determine the protective effect of the pineal gland hormone against the DNA damage induced by cyclophosphamide (CP), an anti-tumor agent that is widely applied in clinical practice. DNA damage was induced in rats by a single intraperitoneal injection of CP (20 or 50 mg/kg). Animals received melatonin during the dark period for 15 days (1 mg/kg in the drinking water). Rat bone marrow cells were used for the determination of chromosomal aberrations and of formamidopyrimidine DNA glycosylase enzyme (Fpg)-sensitive sites by the comet technique and ofXpf mRNA expression by qRT-PCR. The number (mean ± SE) of...

Genotoxicity, anti-melanoma and antioxidant activities of Hymenaea courbaril L. seed extract Anais da ABC (AABC)
SPERA,KAMILLE D.; FIGUEIREDO,PATRÍCIA A.; SANTOS,PAMELA C.E; BARBOSA,FERNANDO C.; ALVES,CAIO P.; DOKKEDAL,ANNE LÍGIA; SALDANHA,LUIZ LEONARDO; SILVA,LUCIANA P.; FIGUEIREDO,CARLOS ROGÉRIO; FERREIRA,PAULO CESAR; SILVA,REGILDO M.G. DA.
Abstract: Hymenaea courbaril has been used to treat different diseases, although its properties are yet to be scientifically validated. The objective of this study was to determine the cytotoxicity, genotoxicity, antigenotoxicity and antioxidant potentials of hydroethanolic extract from H. courbaril seeds. Therefore, for the cytotoxicity test an anti-melanoma assay was performed in B16F10 strain cells. The genotoxicity and antigenotoxicity was evaluated in bone marrow cells (Permit number: 002/2010) of mice, the antioxidant activity was determined by the DPPH test and the total flavonoid content was also determined. The hydroethanolic extract showed antigenotoxic effect and antioxidant activity. It was verified that total flavonoid content was 442.25±18.03...
